Title : Quantitative shotgun proteomics to solve identity issues of human regulatory T cells

Much interest exists in development of therapeutic strategies to manipulate regulatory T cells (Tregs) in inflammatory disorders or for treatment of cancer. Such efforts are complicated by the close relationship between Tregs and conventional CD4 T cells (Tconv) and by instability of certain Treg populations. To find discriminatory properties, we have used high throughput label-free quantitative shotgun proteomics and transcriptomics on primary and in vitro expanded populations of human Tregs and Tconv. We found novel surface markers that mark cells with specific functional properties. Furthermore, we discovered signaling adaptations in Tregs, which allow inflammatory cytokines to mobilize select functions in Tregs (e.g. homing capacity) without eliciting undesirable production of cytokines, such as IFNγ and IL-2. Importantly, most of these findings could not have been made on the basis of isolated transcriptome analysis, underscoring the power of direct proteome measurement.
