Cellular and molecular immunology of chronic Inflammatory diseases - Guy GOROCHOV

Team Research program

Our team played a pioneering role in the definition of human regulatory cells and their study in chronic inflammatory disorders such as Lupus, Scleroderma, Sarcoidosis and also graft rejection/tolerance.
We recently focused on the study of the balance between effector and regulatory T cells and on their relations with pathogens and commensals, and very recently developed a platform dedicated to study the interface between adaptive immunity and gut microbiota. Immune microbiota commensalism is now being studied in Multiple Sclerosis patients, but also in cases of immunodeficiencies associated with autoimmunity.

Technical expertise

In vitro molecular evolution of proteins using phage display, Antibody and TCR engineering, Deep sequencing and analysis of immune repertoires, Single cell PCR.

Main achievements

  • First generation of Chimeric Antigen Receptors (CARs) under supervision of Pr. Zelig Eshhar 1989).
  • First description of Pseudo-Lymphomas with anti-HIV specificity (Journal of Clinical Investigation 1996).
  • First quantitative measurement of global T cell repertoire perturbation, applied to the demonstration of repertoire reconstitution in HIV treated patients (Nature Medicine 1997).
  • n vitro molecular evolution of chemokines as HIV antivirals (Journal of Virology 2003, PNAS 2008).
  • Treg amplification and exhaustion explain immune paradox of sarcoidosis (Journal of Experimental Medicine 2006)
  • Novel phenotypic definition of human Tregs (Immunity 2009)
  • A novel biomarker for identifying effector Treg cells (European Patent application Nb. 13306106.9 2013).
